Altered Transcriptional Regulation

چکیده

The molecular mechanism involved in altered regulation of the rate-limiting enzyme in hepatic gluconeogenesis, phosphoenolpyruvate carboxykinase (PEPCK), during endotoxemia is not completely understood. We examined, therefore, the effect of a nonlethal dose of Escherichia coli endotoxin on PEPCK gene expression in fasted rats. 5 h after endotoxin treatment, the PEPCK transcription rate and the amount ofmRNAPEI'K were significantly decreased at a time when the insulin/glucagon (I/ G) molar ratio and plasma corticosterone levels were significantly increased. Similar results were observed in a time course study, in which altered cAMP induction of PEPCK gene expression paralleled changes in the I/G molar ratio. In diabetic rats treated with endotoxin, PEPCK gene expression was decreased in the absence, however, of an increased I/G molar ratio. This finding indicates that other factors, such as inflammatory mediators or cytokines, alter PEPCK gene transcription during endotoxemia. IL-6, a putative mediator of endotoxin action in the liver, had no effect on PEPCK gene expression in fasted rats, but did decrease cAMP induction ofPEPCK gene expression. These results indicate that, during endotoxemia, regulation of PEPCK gene expression is influenced by inflammatory mediators in addition to the classical endocrine hormones. IL-6, however, does not appear to be involved directly in the altered regulation ofthe PEPCK gene during endotoxemia. (J. Clin. Invest. 1991. 88:811-816.)

Regulation of miR159 and miR396 mediated by Piriformospora indica confer drought tolerance in rice

Drought stress is one of the most determinative factors of agriculture and plays a major role in limiting crop productivity. This limitation is going to rising through climate changes. However, plants have their own defense systems to moderate the adverse effects of climatic conditions. MicroRNA-mediated post-transcriptional gene regulation is one of these defense mechanisms. The root endophyti...

I-34: Steroid Hormone Signalling at the FetomaternalInterface

Background: Progesterone is indispensable for differentiation of human endometrial stromal cells (HESCs) into decidual cells, a process that critically controls embryo implantation. However, HESCs also abundantly express androgen receptors (AR), yet the role of this member of the superfamily of ligand-dependent transcription factors in the decidual process remains poorly elucidated. Linking CREB function with altered metabolism in murine fibroblast-based model cell lines

The cAMP-responsive element binding protein CREB is frequently overexpressed and activated in tumors of distinct histology, leading to enhanced proliferation, migration, invasion and angiogenesis as well as reduced apoptosis. The de-regulated expression of CREB might be linked with transcriptional as well as post-transcriptional regulation mechanisms. We show here that altered CREB expression l...

Potential roles of 5´ UTR and 3´ UTR regions in post-trans-criptional regulation of mouse Oct4 gene in BMSC and P19 cells

Objective(s):OCT4 is a transcription factor required for pluripotency during early embryogenesis and the maintenance of identity of embryonic stem cells and pluripotent cells. Therefore, the effective expression regulation of this gene is highly critical. UTR regions are of great significance to gene regulation. In this study, we aimed to investigate the potential regulatory role played by 5´UT...
